Which role should a new player take?

Rotate roles

The lobby has a Random Role button (rolling it three times unlocks the Indecisive Monkeys achievement, thirty times unlocks Rollaholic). Swapping after a few levels is the fastest way to understand what your teammates actually need from you; several launch reviews say the game only clicked once they had played all three seats.

Can two people play, or one?

No. The store page says "exactly three monkeys required" and the developer confirmed on the Steam forum that every player needs a copy. Use the lobby browser to fill a third seat.
